Safety Concerns and Case Reports
Two recent case reports have raised safety concerns about Melanotan II use. A 2026 case report documented changes in oral mucosa associated with Melanotan II injections, while a 2025 study in the International Journal of Oral and Maxillofacial Surgery examined whether Melanotan II nasal spray might be a risk factor for oral mucosal malignant melanoma. These reports suggest potential serious adverse effects, though the causal relationships remain unclear from individual case studies.
Animal Research
A 2024 study published in the European Journal of Pharmacology examined the effects of Melanotan II in male rats using a chronic unpredictable stress model. The researchers found antidepressant-like and antistress effects from this ACTH(4-10) synthetic analog, comparing it with another compound called Semax. However, animal studies may not translate directly to human effects.
Additional animal research from 2024 explored metabolic effects, with studies examining the role of melanocortin receptors in feeding regulation and metabolic signaling pathways. These provide background on the biological mechanisms but don't establish clinical efficacy or safety in humans.
Technical and Forensic Studies
Recent publications have also addressed detection methods for Melanotan II (referred to as "Barbie drug" in forensic contexts) and chemical synthesis techniques for creating modified versions of the compound. A 2024 study recommended Melanotan II as a tool compound for melanocortin receptor research.
Clinical Trial
One active clinical trial (NCT07437560) is currently recruiting participants to study Melanotan II as an adjunct to narrowband UV-B phototherapy for vitiligo repigmentation. This Phase 2 trial represents the most rigorous ongoing research into therapeutic applications.
Evidence Limitations
The current evidence base is notably thin, consisting primarily of case reports, animal studies, and basic research rather than robust clinical trials. The safety profile in humans remains poorly characterized, and efficacy data from controlled human studies is extremely limited.
