Celiac Disease

The most established clinical application for larazotide is in celiac disease management. However, a subsequent Phase 3 efficacy and safety trial (NCT03569007) aimed at evaluating larazotide for relief of celiac disease symptoms was terminated, suggesting challenges in demonstrating clinical efficacy at this stage of development.

Recent research indicates larazotide may help protect the intestinal mucosal barrier, which is compromised in celiac disease. The compound appears to work through various cellular mechanisms to maintain barrier function, though clinical translation of these effects remains under investigation.

Long COVID

A Phase 2 trial (NCT05747534) is currently investigating larazotide acetate (AT1001) for treating Long COVID, though this study is no longer actively recruiting participants. The rationale for this application likely relates to the compound's potential effects on barrier function and inflammation.

Post-COVID Multisystem Inflammatory Syndrome

Emerging research has explored larazotide's use in children with post-COVID multisystem inflammatory syndrome, with one study reporting effects on viral spike antigen clearance and recovery, though more evidence is needed to establish clinical efficacy.
