Reproductive and Pubertal Development
Several studies have investigated kisspeptin's role in pubertal timing and reproductive function. A 2026 study examined serum kisspeptin and DLK1 profiles during GnRH stimulation in children with central precocious puberty compared to those with premature thelarche, providing insights into diagnostic biomarkers. Another study explored how stress affects the KNDy-GnRH pulse generator system, linking HPA-axis activation to female reproductive dysfunction through neuroendocrine mechanisms.
Research has also examined kisspeptin's involvement in pregnancy complications, with one study investigating how the progesterone-KISS1 axis may impair amniotic epithelial cell function and contribute to premature rupture of membranes.

Clinical Trials
Multiple clinical trials have been completed or are ongoing. A Phase 3 trial (NCT03286517) investigated the relationship between kisspeptin signaling sensitivity and pubertal onset in boys. Currently recruiting studies include a Phase 1 trial (NCT07224490) quantifying GnRH neuronal function and an observational study (NCT06545994) tracking serum kisspeptin levels throughout pregnancy in women with PCOS.
The research demonstrates kisspeptin's expanding therapeutic potential while highlighting the need for larger clinical studies to establish efficacy across different indications.
