Clinical Applications of Lenalidomide
Lenalidomide is primarily used in the treatment of various hematological malignancies, with substantial evidence supporting its efficacy in multiple myeloma and certain lymphomas.
Multiple Myeloma:
Lenalidomide is a key component in the treatment regimen for multiple myeloma, both newly diagnosed and relapsed/refractory cases. A study published in the Annals of Medicine (2026) investigated lenalidomide in combination with selinexor, bortezomib, and dexamethasone for newly diagnosed multiple myeloma patients with high-risk factors. This combination is part of a strategy to improve outcomes in high-risk patients. Additionally, lenalidomide is often used as maintenance therapy post-autologous stem cell transplantation. Lymphomas:
Lenalidomide is also used in various lymphomas, including follicular lymphoma and MYC-associated B-cell lymphomas. Myelofibrosis Complicated with Multiple Myeloma:
There is a notable case of lenalidomide inducing the disappearance of specific chromosomal abnormalities (del(7q)) in patients with myelofibrosis complicated by multiple myeloma, as illustrated in a 2026 case report from The Japanese Journal of Clinical Hematology.
Overall, lenalidomide plays a significant role in the management of multiple myeloma and some lymphomas, with ongoing research exploring additional applications and optimal combinations. As research continues, its use may be refined to maximize patient outcomes.
